| The cryopreservation of ovarian tissues is a technology with significant potential for the preservation of the genetic resource materials of working dogs, including guide dogs for the blind. However, no attempt has been reported on cryopreservation of the canine ovary. Thus, we evaluated a vitrification method for cryopreservation of canine ovaries and determined the potential functionality of vitrified-warmed canine ovaries by means of transplantation into non-obese diabetic-severe combined immunodeficiency (NOD-SCID) mice. | Chondroitinase ABC (C-ABC) is expected to be a novel agent for chemonucleolysis. The effect of C-ABC was investigated by magnetic resonance (MR) and radiograph. C-ABC was administered into the lumbar intervertebral disks on the clinically normal beagles (n=5), in a dose of 50 μl (12.5 units as C-ABC). MR scans were performed pre-dose, and 1, 3, 7, 14 and 28 days after administration of C-ABC, and the signal intensity (SI) of the nucleus pulposus was measured. | To estimate the maternal effects of dog breeds using mitochondrial DNA(mtDNA) haplotypes in the dogs with several clinical disorders, 600 base pairs of mtDNA D-loop region were amplified from 365 dogs and were determined for mtDNA sequences. The diversity of the 600-bp sequences was classified into 64 haplotypes, including 46 newly discovered haplotypes, and the haplotypes were grouped into four clusters I to IV. Lineage analysis using the mtDNA haplotype indicated that each dog breed genetically comprises one or a few mtDNA haplotypes. | Growth of the tumor autoinduced by 20-methylcholanthrene (MC) in rats was inhibited after administration of Toxoplasma lysate antigen (TLA). The antitumor activity of TLA was most obvious in the early stage of tumoral growth. When TLA was administered to rats before the appearance of tumor, tumor formation was delayed slightly. Histopathological studies revealed dense growths of spindle tumor cells in untreated control rat, while enlarged central necrosis with the infiltration of lymphocytes and neutrophils was apparent in TLA-treated rats. | To obtain B-mode ultrasound images of mammary glands in dairy heifers at different stages of growth, 25 clinically normal Holstein heifers were used. The heifers were divided into 5 groups (n=5/group) by stage of their growth: 2-month-old (group 1), 5-month-old (group 2), postpuberty (group 3), mid (group 4), and late (group 5) pregnancy. Furthermore, the sections of mammary glands were observed grossly at postmortem examination in one heifer in each group. Ultrasound images varied with the development of mammary glands. | A 9-month-old steer was autopsied due to recurrent ruminal tympany. A macroscopic examination found an enlarged caudal mediastinal lymph node, and a section of the lymph node revealed necrosis with marked calcification, similar to tuberculous lymphadenitis. Histopathologically, the lesion consisted of multiple coagulative necrotic foci and fibrosis with macrophage, lymphocyte, eosinophil and multinucleated giant cell infiltration. Non-uniform width hyphae were detected in the necrotic area and within the cytoplasm of the multinucleated giant cells, and they were found to be anti-Rhizopus arrhizus antibody positive in an immunohistochemical examination. | In order to identify mechanisms responsible for the anti-tumor effects of Toxoplasma lysate antigen (TLA), we used an in vitro 51Cr release assay to study the functional properties of plastic-adherent cells during induction of splenic cytotoxic activity by TLA. Cytotoxic activity of non-adherent cells was measured in all experiments after a 6 days incubation. Induction of cytotoxic non-adherent cells by TLA required the presence of plastic-adherent spleen cells. In contrast, rhIL-2 alone was able to induce transformation of cytotoxic non-adherent cells from non-adherent spleen cells. | Spleen cells from Toxoplasma lysate antigen (TLA)-sensitized BALB/c mice showed the strong cytotoxic activity against both natural killer (NK)-sensitive cells (YAC-1 and RL♂-1) and NK-insensitive cells (P-815), when incubated with TLA or recombinant human IL-2 (rhIL-2). The increment of TLA concentration in culture medium increased the cytotoxic activity. | 三次元超音波検査法の小動物臨床への応用の可能性を探求するために，妊娠後期の6 歳 のビーグル犬に対して，腹部用三次元プローブを用いた三次元超音波検査法を出産まで数 日毎に行い，8 頭の胎子の顔を描出しようと試みた。胎子の顔の三次元超音波画像が全ての 検査日において少なくとも1 頭の胎子で得られたが，胎子が成長するにつれて良好な画像 を得るのは難しくなった。交配後43 日目ならびに45 日目では胎子の顔の鮮明な三次元画 像が容易に得られた。技術的に解決しなければならない問題がいくつか残されているもの の，適切に用いさえすれば，三次元超音波検査法は従来の二次元超音波検査法を補足する 方法として小動物臨床に応用可能であると考えられる。 This study was delineated to explore the possibility of using three-dimensional ultrasonography in small animal practice.
